Studies on Changes in Serum Protein Content and Its Electrophoretic Pattern in Rabbits Intoxicated with Carbone Tetrachloride


Abstract
Proteins are fundamental constituents of all protoplasm and involved in the structure of the living cell and in function for the latter is brought about by enzymes which are themselves proteins.
In normal individuals approximately 0. 2~0.3 gm of plasma protein is formed daily per Kg. of body weight.. Also, functions of the plasma proteins are ; 1) to maintain osmotic pressure in blood, 2) to act as a reserve of protein for tissue repair and growth, 3) to act as pH buffers, 4) to act as transport of lipid, bilirubin, vitamin A,D, and E, steroid hormone, 5) to act as immunlogical agents, 6) to provide factors necessary for normal blood coagulation, 7) to provide necessary enzymes in the blood.
Protein synthesis is now believed to be a function of the ribosomes and the associated membranes of the endoplasmic reticulum. The first indication of a relationship between ribosome and protein synthesis was presensed by Casperson and Brachet.
Keller. et al and Mirsky. et al demonstrated in vivo most rapid and early incorporation of the amino acids into the microsomal porsion of fractioned cells, suggesting this site as a major one of intracellular protein formation.
Since the rough endoplasmic reticulum is believed currently to participate in protein synthesis, the alterations produced by carbon tetrachloride suggested that there might be a defect in this process.
Changes of rabbit¢¥s serum protein and its electrophoretic pattern on acute CCI4 intoxication was experimentally invesigated. Ten adult male rabbits weighing 1.8 to 2.0kg. were used throughout investigation.
Blood samples was drawn from heart 5ml. and serum level of total protein, albumin, globulin and protein electrophoresis were tested at the interval of the 1st, 7th, 15th hour and 1st. 2nd, 3rd, 5th, 8th, 12th day after injection of a lml of 30% CC14 in olive oil into subcutis.
The following result were obtained:
1) Normal control serum protein, albumin and globulin were 6.01¡¾ 0.32gm/dl, 3.70¡¾
